بررسی اثر سمیت نانوذرات اکسید روی سنتز شده به روش سبز با عصاره گیاه خرنوب بر رده سلول سرطانی پستان MDA-MB231 و ارزیابی اثرات ضد رگزایی آن

چکیده
مقدمه

سنتز نانوذرات فلزی به روش سبز یکی از روش‌­های کارامد دارای حداقل عوارض جانبی در درمان سرطان به شمار می­‌رود. هدف از مطالعه حاضر بررسی سمیت سلولی نانوذرات اکسید روی سنتز شده با عصاره خرنوب بر سلول­‌های سرطان پستان و بررسی اثر ضد رگ­زایی این نانوذرات در مدل پرده کوریوالانتوئیک جوجه می‌­باشد.

مواد و روش‌­ها

در این پژوهش سلول­‌های سرطانی MDA-MB231 به مدت 24 ، 48 و 72 ساعت تحت تیمار با غلظت­‌های مختلف (31/2 -250 میکرو گرم بر میلی لیتر) از نانوذرات قرار گرفته و درصد زیستایی سلول­‌ها با استفاده از تست MTT بررسی شد. همچنین با انجام تست CAM روی تخم‌­مرغ­‌های نطفه‌­دار اثر ضد رگ‌­زایی این نانوذرات با اندازه‌­گیری تعداد رگ، طول عروق، طول و وزن جنین بررسی شد.

یافته­‌ها

نانوذرات اکسیدروی سنتز شده به روش سبز به صورت وابسته به دوز و زمان باعث القای سمیت سلولی بر روی رده سلول­‌های سرطان پستان می­‌شود (0/001>P). میزان IC50 در این سلول‌­ها 62/5 میکرو گرم بر میلی لیتر بدست آمد. بررسی میزان رگ‌­زایی در مدل پرده کوریوالانتوئیک نشان داد که نانوذرات اکسیدروی، با توجه به دوز، به صورت معنی­‌داری منجر به کاهش تعداد عروق (0/001
P) ;(0/01 P) و طول عروق (0/05P) ;( 0/001P ) و قد جنین (0/05P ) و وزن جنین ((0/05P در دوز بالا) می­‌گردد.

نتیجه‌­گیری

نانوذرات اکسیدروی سنتز شده با عصاره خرنوب با القای اثر کشندگی و اثرات ضد رگ‌­زایی بر سلول‌­های سرطان پستان می‌­تواند به عنوان گزینه مناسبی در درمان سرطان مورد توجه قرار گیرد.

Introduction

Green biosynthesis of the metallic nanoparticles is considered as one of the effective method for cancer treatment with minimum side effects. The objective of this study was the evaluation of cytotoxicity of zinc oxide (ZnO) nanoparticles synthesized with Ceratonia siliqua extract on the breast cancer cells and assessment of anti-angiogenesis properties of these nanoparticles in the chorioallantoic model (CAM).

Methods & Materials

In this investigation, the MDA-MB231 cancer cells line was treated for 24, 48 and 72 hours at different concentrations of ZnO nanoparticles (31.2 to 250 µg/ml) synthesized with Ceratonia siliqua extract and the percentage of cell viability was evaluated using an MTT assay. In addition, to determine the anti-angiogenesis effect, Ross Fertilized Chicken eggs were used in the CAM assay. After 12 days, the vessel number, vessels length, embryo length, and weight were measured.

Results

The findings showed that the green synthesized ZnO nanoparticle induces a cytotoxicity effect on breast cancer cell line (P≤ 0.001) depended in dose and time. The IC50 values were found about 62.5 μg/ml.

Evaluation of angiogenesis in the CAM model showed that ZnO nanoparticles in the dose ratio used, significantly reduced the number of vessels (P≤0.01); (P≤0.001) and the length of the vessels (P≤0.001); (P≤0.05) and embryo size (P≤0.05) and embryo weight ((P≤0.05) only in the high-dose).

Conclusion

ZnO nanoparticles synthesized with Ceratonia siliqua extract having a cytotoxic effect on breast cancer cells and anti-angiogenic effect can be an appropriate candidate in cancer treatment.
