تأثیر هشت هفته تمرین هوازی و مصرف قهوه سبز بر بیان ژنABCG1 لنفوسیت، Apo-B100 و برخی از شاخص‌های خطر‌زای قلبی و عروقی زنان غیر ورزشکار دارای اضافه وزن

چکیده
مقدمه

بیماری­های قلبی­عروقی و آتروسکلروزیس مهمترین عامل مرگ و میر در کشورهای صنعتی هستند. هدف از این مطالعه بررسی تأثیر هشت هفته تمرین هوازی و مصرف قهوه سبز بر بیان ژنABCG1 لنفوسیت، Apo-B100 و برخی از شاخص­های خطر­زای قلبی­عروقی زنان غیر ورزشکار دارای اضافه وزن بود.

مواد و روش­ها

در این پژوهش نیمه تجربی کارآزمایی بالینی یک­سوکور، به­طور تصادفی30 نفر بانوان میانسال دارای اضافه وزن تمرین نکرده­ باشگاه ایروبیک مشهد، با رضایت­نامه کتبی انتخاب و به دو گروه 15 نفری تمرین هوازی (8 هفته، 3 جلسه در هفته، زمان از 20 تا 60 دقیقه در هفته آخر) همراه با مصرف روزانه قهوه سبز و مصرف دارونما تقسیم شدند. خون­گیری صبح ناشتا، قبل از شروع تمرینات و 48 ساعت بعد از آخرین جلسه تمرین انجام شد. جداسازی لنفوسیت­ها با استفاده از محلول فایکول و بررسی بیان ژن با روش RT-PCR و اندازه­گیریApo-B100 با روش الایزا انجام شد. داده­ها با استفاده از آزمون ANCOVA و نرم­افزار SPSS-22 تجزیه و تحلیل شدند (p<0.05).

یافته‌ها

نتایج نشان داد که تمرین هوازی باعث افزایش معنی­دار بیان ژن ABCG1، کاهش معنی­دار Apo-B100 و کاهش معنی­دار نسبت LDL/HDL و CT/HDL در هر دو گروه شد (0001/0p=). اما تمرین هوازی همراه با مصرف قهوه سبز فقط بر کاهش Apo-B100 (028/0p=، 532/5f= ) تأثیر معنی­دار داشت.

نتیجه­گیری

نتایج نشان داد که چون تمرین هوازی همراه با مصرف قهوه سبز باعث افزایش بیان ژن ABCG1 و کاهش Apo-B100 می­شود شاید بتواند نقش مؤثری درافزایش RCT و پیشگیری از آترواسکلروز و بیماری­های قلبی­عروقی داشته باشد.

Introduction

Cardiovascular diseases and atherosclerosis are the most important cause of death in industrialized countries. The aim of this study was the investigation of effect of eight weeks aerobic exercise and consumption of green coffee on gene expression ABCG1, Apo-B100 and some of the Cardiovascular risk factors in untrained and overweight women.

Materials and Methods

In this quasi-experimental single blind study, 30 untrained overweight middle-aged women in Mashhad Aerobic Club, with written consent, were randomly divided to two groups of aerobic exercises (8-Weeks, 3days/Week, 20 min/day, at the last week 60 min/day) with green coffee (N=15) and placebo (N=15) daily consumption. Blood samples were collected in the morning fasting before exercise and 48 hours after the last training session. Isolation of lymphocytes using Ficoll solution and for gene expression was analyzed by RT-PCR and Apo-B100 were measured by ELISA method. The data were analyzed by ANCOVA and SPSS-22 software (p <0.05).

Results

The results showed that aerobic exercise significantly increased the ABCG1 gene expression and significant reduction of Apo-B100 and LDL/HDL and CT/HDL ratio in both groups (p = 0.0001). But the aerobic exercise combined with green coffee consumption had only significant effect on Apo-B100 reduction (p= 0.028, f= 5.532).

Conclusion

The results showed that aerobic exercise combined with green coffee consumption increased ABCG1 gene expression and decreased Apo-B100 may have an effective role in increasing RCT and preventing atherosclerosis and cardiovascular disease.
