تأثیر یک دوره تمرین ترکیبی بر بیان ژن ABCB4 گلبول‌های سفید و نسبت HDL به LDL در مردان میانسال بعد از انجام عمل بای‌پس عروق قلبی

چکیده
مقدمه

بیماری‌های قلبی و عروقی در سراسر جهان و از جمله ایران شیوع زیادی دارند و در این بین تصلب شرایین عامل عمده مرگ و میر در تمام دنیا محسوب می‌شود. هدف پژوهش حاضر بررسی تاثیر یک دوره تمرین ترکیبی بر بیان ژن ABCB4 و نسبت HDL به LDL در مردان میانسال بعد از انجام عمل بای‌پس عروق قلبی می‌باشد.

مواد و روش­ها

نمونه آماری این تحقیق را 30 مرد میانسال تشکیل دادند که قبلاً عمل بای‌پس عروق کرونری انجام داده بودند( بیماران CABG). این تحقیق از نوع نیمه تجربی است که به روش نمونه گیری انتخابی به‌طور تصادفی در دو گروه کنترل(15نفر), تمرین ترکیبی استقامتی و مقاومتی، 15 نفر) قرار گرفتند. میانگین وزن آزمودنی‌ها 5.87± 75.45 (کیلوگرم) و شاخص توده بدن 56/1± 11/25 (وزن×قد²) بود. قبل و بعد جلسه تمرینی (24 جلسه ‌3 روز در هفته) از آزمودنی‌ها خون‌گیری به عمل آمد. پس از جداسازی گلبول‌‎های تک هسته‌ای با استفاده از فایکول و تخلیصmRNA تغییرات بیان ژن به‌ روش Real-Time PCr انجام شد. داده‌ها با آزمون آماری تحلیل واریانس با اندازه‌های تکراری تجزیه و تحلیل شد.

یافته‌­ها

هشت هفته تمرین منجر به افزایش معنی‌داری در بیان ژن ABCB4 و کاهش معنی‌دار سطوح پلاسمایی کلسترول تام گروه تمرین ترکیبی در مقایسه با گروه کنترل شد.

نتیجه‌­گیری

به نظر می‌رسد تمرین ترکیبی به عنوان بخشی از مراحل بازتوانی قلبی باعث افزایش بیان ژن ABCB4 و انتقال معکوس کلسترول و کاهش سطوح پلاسمایی کلسترول تام و بهبود شرایط آندوتلیالی دربیماران مرد میانسال بعد از انجام عمل بای‌پس عروق قلبی می‌شود.

چکیده English

Introduction

Cardiovascular diseases are very common all over the world, including Iran. Meanwhile, atherosclerosis is the leading cause of death worldwide. The aim of this study was to investigate the effect of combined training on ABCB4 gene expression and HDL to LDL ratio in middle-aged men after coronary artery bypass graft.

Materials and Methods

The statistical population consisted 30 middle-aged men that had previously undergone coronary artery bypass graft. This research is quasi-experimental which by random sampling method, they were randomly divided into two groups of control (n=15) and combined training (endurance and resistance, n=15). Blood samples were taken from the two groups before and after the training session. After isolation mononuclear cells by using follicles and mRNA purification, gene expression variation was performed by Real- Time PCR. Data were analyzed by repeated measures variance analysis with SPSS software version 16.

Results

Eight weeks of training resulted in a significant increase in ABCB4 gene expression and significant reduction in plasma total cholesterol levels in the combined training group compared to the control group.

Conclusion

Combined training, as part of cardiac rehabilitation in patients performing coronary artery bypass graft by effecting on ABCB4 gene expression, involved in fat metabolism, improves reverse cholesterol transfer process.
