نقش آنتی بیوفیلمی تریکلوزان بر جدایه های استرپتوکوکی واجد سورتاز A در کنترل پوسیدگی دندان

چکیده
مقدمه

یکی از رایجترین مشکلات دوران کودکی، پوسیدگی دندان بوده و استرپتوکوکوس موتانس رایجترین باکتری عامل عفونت دندان است. این مطالعه با هدف بررسی اثر تریکلوزان بر تشکیل بیوفیلم ناشی از فعالیت آنزیم سورتازA استرپتوکوکی در کنترل پوسیدگی دندان انجام شد.

مواد و روش­‌ها

در این مطالعه تجربی، 150 نمونه از سطح بوکال و لینگوال دندان‌های خلفی دانش‌آموزان دبستانی در گرگان، طی سال 1398 گرفته شد. سویههای استرپتوکوکوس موتانس با استفاده از تست‌های میکروبیولوژیک و بیوشیمیایی شناسایی شدند. ژن SrtA با استفاده از واکنش زنجیرهای پلیمراز (PCR) با آغازگرهای اختصاصی شناسایی شد. حداقل غلظت مهاری تریکلوزان بر جدایهها به روش میکرودایلوشن براث تعیین شد. تشکیل بیوفیلم و اﺛﺮات ﺿﺪ ﺑﯿﻮﻓﯿﻠﻤﯽ تریکلوزان به روش ﻣﯿﮑﺮوﺗﯿﺘﺮ ﭘﻠﯿﺖ اﺻﻼح ﺷﺪه ﺑﺮرﺳﯽ ﺷﺪ.

یافته‌­ها

فراوانی استرپتوکوکوس موتانس 3/15 درصد بود که 87٪ آن‌ها قادر به تولید بیوفیلم بودند. 19 جدایه (6/82%) واجد ژن srtA شناسایی شد. حداقل غلظت مهاری تریکلوزان که رشد50% جدایه‌ها را مهارکرد (MIC50 8 میکروگرم بر میلی‌لیتر بود که 8 برابر کمتر از غلظت لازم برای مهار رشد 90 درصد جدایه‌ها بود (MIC90=64μg/ml)، به طوری که 90% جدایه‌ها فاقد توانایی تشکیل بیوفیلم در غلظت MIC بودند.

نتیجه‌­گیری

نتایج نشان داد که بین حضور سورتاز A و تشکیل بیوفیلم باکتریایی ارتباط وجود دارد و تریکلوزان اثر ضد بیوفیلمی خوبی را در شرایط آزمایشگاهی نشان داد.

The Anti-Biofilm Role of Triclosan on the Activity of Streptococcal Isolates containing Sortase A in Control of Dental Caries

Introduction

One of the most common childhood problems is tooth decay and Streptococcus mutans is the most common bacterium that causes the infection. The purpose of the study was to investigate the effect of triclosan on biofilm formation due to streptococcal sortase A enzyme activity in the control of dental caries.

Materials and Methods

In this experimental study, 150 samples were taken from the buccal and lingual surfaces of posterior teeth of pupils in Gorgan in 2019. S. mutans strains were identified using conventional microbiological and biochemical tests. The SrtA gene was identified using polymerase chain reaction (PCR) with specific primers. The minimum inhibitory concentration (MIC) of triclosan against the isolates was determined by the broth microdilution method. Biofilm formation and antibiofilm effect of triclosan were assessed using the modified microtiter plate assay

Results

The frequency of S. mutans was 15.3%, of which, 87% were capable of biofilm production. 19 isolates (82.6%) with the srtA gene were identified. The concentration of triclosan, that inhibited 50% growth of isolates (MIC50) was 8μg /ml, which was 8-fold less than the concentration required to inhibit the growth of 90% of isolates (MIC90=64μg/ml), so that 90% of the isolates lacked the ability to form biofilms at MIC concentration (P<0.05).

Conclusion

The results showed that there is a relationship between the presence of sortase A and the formation of bacterial biofilm and triclosan showed a good anti-biofilm effect in vitro condition.
